TROP2-targeted anti-Frizzled bispecific antibody is an experimental bispecific antibody developed by Genentech to selectively inhibit the Wnt signaling pathway in tumor cells. The antibody is designed to address the dose-limiting intestinal toxicities typically associated with systemic Wnt inhibition, which occur because Wnt signaling is essential for normal intestinal homeostasis. By utilizing TROP2 (Trophoblast cell-surface antigen 2) as a tumor-selective anchor—a receptor highly expressed in various epithelial cancers but absent in Wnt-dependent normal intestinal stem cells—the antibody selectively delivers a potent Wnt-inhibitory signal to tumor tissues. It simultaneously binds to TROP2 and Frizzled (FZD) receptors, effectively blocking oncogenic Wnt signaling and suppressing tumor growth in preclinical models of colorectal cancer (CRC) and pancreatic ductal adenocarcinoma (PDAC) with minimal impact on normal intestinal tissue.
