TROP2 targeting antibody-drug conjugates are a class of targeted cancer therapies that combine a monoclonal antibody specific for Trophoblast cell surface antigen 2 (TROP2) with a cytotoxic drug (payload), connected by a chemical linker. TROP2 is a cell surface glycoprotein highly expressed in various solid tumors, including breast cancer, and is implicated in tumor progression, metastasis, and resistance to therapy[1][3]. The antibody component specifically binds to TROP2 on tumor cells, allowing for the internalization and intracellular release of the cytotoxic payload, which induces cell death with minimal effect on normal tissues. The most clinically advanced and representative TROP2-targeting ADCs include sacituzumab govitecan and datopotamab deruxtecan, both of which are approved or in late-stage development for metastatic breast cancer and other TROP2-expressing tumors[1][2][3].
