TROP2-targeting T-cell engagers (TCEs) are a class of bispecific immunotherapies designed to treat solid tumors by simultaneously binding to Trophoblast cell-surface antigen 2 (TROP2) on cancer cells and the CD3 epsilon subunit of the T-cell receptor complex on T-cells. This dual binding facilitates the formation of a cytolytic synapse, leading to the activation and redirection of T-cells to eliminate TROP2-expressing malignant cells independently of MHC-I presentation. TROP2 is highly expressed in various epithelial cancers, including breast, lung, and urothelial carcinomas, making it a high-priority target for redirected T-cell therapies. Several candidates are currently in early-phase clinical trials, exploring different formats such as IgG-based bispecifics or smaller antibody fragments.
