TROP2-TRACTr is a next-generation tumor-activated T cell engager (TCE) engineered to target **trophoblast cell surface antigen 2 (TROP2)**, a cell-surface glycoprotein overexpressed in a wide array of solid tumors including breast, lung, and bladder cancers. It is being developed by Janux Therapeutics as part of their modular TRACTr platform. Unlike traditional TROP2-targeted therapies such as antibody-drug conjugates (ADCs), TROP2-TRACTr is designed to achieve potent anti-tumor activity even at low TROP2 expression levels, while minimizing toxicity to healthy tissue. The molecule demonstrated strong preclinical efficacy and an improved safety profile (no cytokine release syndrome or healthy tissue toxicity) compared to earlier TCEs and ADCs, positioning it as a potentially best-in-class TROP2-targeted therapy for solid tumors[1][2][3].
