Overview

Tropoflavin (7,8-dihydroxyflavone) is a naturally occurring flavonoid that acts as a potent and selective small-molecule agonist of the tropomyosin receptor kinase B (TrkB), the primary signaling receptor for brain-derived neurotrophic factor (BDNF). Originally identified in plants such as *Godmania aesculifolia*, *Tridax procumbens*, and *Primula vulgaris*, it is orally bioavailable and capable of penetrating the blood-brain barrier, making it a significant research tool and potential therapeutic for various neurological and psychiatric disorders. Preclinical studies have demonstrated its efficacy in animal models of Alzheimer's disease, Parkinson's disease, depression, and traumatic brain injury. While it is currently sold as an over-the-counter nootropic dietary supplement in the United States, it remains in the preclinical stage of pharmaceutical development for clinical indications. A prodrug derivative, R13, is also under investigation for improved pharmacokinetics in Alzheimer's disease.
