Troriluzole is a third-generation novel prodrug and new chemical entity that acts as a glutamate modulator. It is designed to reduce synaptic levels of glutamate by increasing its uptake from the synapse and decreasing presynaptic release. This mechanism targets excessive glutamatergic activity implicated in neurodegenerative diseases such as spinocerebellar ataxia (SCA), Alzheimer's disease, obsessive-compulsive disorder (OCD), and others. Troriluzole is a tripeptide prodrug of riluzole with improved bioavailability and once-daily oral dosing. Developed primarily by Biohaven Pharmaceuticals, it has shown significant efficacy in slowing disease progression in SCA patients by 50–70% over three years[1][2][4][6].
