Overview

TRP-2 vaccinia virus is a recombinant viral vector vaccine designed for the treatment of melanoma. It utilizes a vaccinia virus platform to deliver the gene for tyrosinase-related protein 2 (TRP-2), also known as dopachrome tautomerase (DCT), which is a melanocyte differentiation antigen frequently overexpressed in melanoma cells. The vaccine is intended to stimulate the host's immune system to recognize and destroy TRP-2-expressing tumor cells through the activation of CD8+ cytotoxic T lymphocytes and natural killer (NK) cells. Preclinical studies have demonstrated that this approach can effectively treat established tumors in mouse models, such as B16 melanoma, while potentially avoiding the widespread autoimmune depigmentation (vitiligo) often seen with other melanocyte-targeted immunotherapies.
