Overview

TRP2-loaded erythrocytes are an investigational cancer immunotherapy consisting of red blood cells (erythrocytes) encapsulated with the melanoma-associated antigen Tyrosinase-related protein 2 (TRP2). Developed using platforms like Erytech Pharma's ERYCAPS, the technology involves loading the antigen into autologous or allogeneic erythrocytes via controlled osmotic lysis and resealing. Upon intravenous administration, these loaded cells are primarily sequestered by the spleen and liver, where they are phagocytosed by professional antigen-presenting cells (APCs). This targeted delivery facilitates the presentation of TRP2 epitopes to T cells, thereby inducing a specific and robust cytotoxic T-lymphocyte (CTL) response against TRP2-expressing tumor cells, such as those in malignant melanoma. This approach aims to enhance the immunogenicity of tumor antigens while leveraging the natural clearance of aged erythrocytes to minimize systemic toxicity.
