Trp2-P30 is an experimental cancer vaccine construct designed to elicit an immune response against melanoma. It consists of a fusion between the tyrosinase-related protein 2 (TRP-2) antigen—a protein highly expressed in melanocytes and melanoma cells—and the P30 epitope, a "universal" T-helper epitope derived from the tetanus toxin. The P30 sequence is included to recruit CD4+ T-cell help, which is critical for the induction and maintenance of a robust CD8+ cytotoxic T-lymphocyte (CTL) response against the TRP-2 self-antigen. This strategy is employed to overcome peripheral tolerance to tumor-associated antigens. The construct has been primarily evaluated in preclinical studies as a DNA vaccine or a recombinant peptide, demonstrating the ability to inhibit tumor growth and prolong survival in melanoma models.
