TRP2-targeted alphavaccine is a viral replicon particle (VRP) cancer vaccine developed by AlphaVax for the treatment of melanoma. The vaccine utilizes a propagation-defective Venezuelan Equine Encephalitis (VEE) virus vector to deliver mRNA encoding the tyrosinase-related protein 2 (TRP2) tumor antigen. TRP2 is a membrane-bound enzyme involved in melanin synthesis that is highly expressed in both normal melanocytes and melanoma cells. Upon administration, the VRPs infect dendritic cells, leading to the expression of TRP2 and the subsequent induction of a potent, antigen-specific T-cell immune response designed to recognize and eliminate melanoma cells. The program has been evaluated in Phase 1 clinical trials to assess safety and immunogenicity in patients with advanced melanoma.
