Drug intelligence / Profile preview
TRPC5 inhibitors are a class of small molecule therapeutics designed to block the transient receptor potential cation channel subfamily C member 5 (TRPC5). TRPC5 is a calcium-permeable, non-selective cation channel expressed in various tissues, including the brain and kidney podocytes. In the kidney, activation of the TRPC5-Rac1 pathway leads to actin cytoskeleton remodeling, podocyte injury, and subsequent barrier breakdown, which manifests as proteinuria. By inhibiting TRPC5, these compounds help stabilize the podocyte actin cytoskeleton, reduce urinary protein loss, and slow the progression of glomerular diseases such as focal segmental glomerulosclerosis (FSGS), diabetic nephropathy, and Alport syndrome. Several TRPC5 inhibitors, such as GFB-887, have entered clinical development for proteinuric kidney diseases.
