TRPH-222 is a novel antibody-drug conjugate (ADC) designed for the treatment of B-cell malignancies such as non-Hodgkin’s lymphoma. It consists of a humanized monoclonal antibody targeting the B-cell surface antigen CD22, site-specifically conjugated via a non-cleavable linker to maytansine—a potent microtubule inhibitor. The SMARTag technology enables precise control over the drug-to-antibody ratio and enhances stability. By binding to CD22 on malignant B cells, TRPH-222 delivers its cytotoxic payload directly into these cells, inhibiting tubulin polymerization and inducing apoptosis. The drug has demonstrated early signs of efficacy and a favorable safety profile in phase 1 clinical trials for relapsed/refractory follicular lymphoma (FL), diffuse large B-cell lymphoma (DLBCL), and mantle cell lymphoma (MCL). It was originally developed by Catalent Biologics using proprietary technology and is being further developed by Triphase Accelerator Corporation[1][2][3][4][5][6][8].
