TRRP@DOX is a preclinical, doxorubicin-loaded targeted redox-responsive peptide drug-delivery system designed for transcatheter arterial chemoembolization of hepatocellular carcinoma. The peptide carrier binds transferrin receptor on tumor-cell surfaces and releases doxorubicin preferentially under the elevated glutathione conditions of tumor cells; doxorubicin then mediates cytotoxicity principally through DNA intercalation and topoisomerase II inhibition. In an orthotopic rat hepatocellular carcinoma model, TRRP@DOX dispersed in lipiodol improved therapeutic outcomes versus a conventional doxorubicin plus lipiodol emulsion.
