This is a combination therapy consisting of three agents used in the treatment of B-cell malignancies, particularly relapsed or refractory chronic lymphocytic leukemia (CLL) and non-Hodgkin lymphoma. TRU-016, also known as otlertuzumab, is a humanized anti-CD37 protein therapeutic that induces direct caspase-independent apoptosis and antibody-dependent cell-mediated cytotoxicity in malignant B cells[1][5]. Idelalisib is an oral small molecule inhibitor targeting phosphoinositide 3‑kinase delta (PI3Kδ), which disrupts signaling pathways critical for the survival and proliferation of malignant B cells[4]. Rituximab is a monoclonal antibody directed against CD20 on B lymphocytes, leading to their depletion through multiple immune-mediated mechanisms. The combination aims to target different antigens and signaling pathways on malignant B cells for synergistic anti-tumor activity. Clinical studies have shown that combinations including these agents can be effective in patients with relapsed or refractory CLL/NHL[1][2][4].
