Overview

TRV-101 is an orally bioavailable, brain-penetrant small-molecule inhibitor of pathological oligomerization of amyloid beta (Aβ) and tau developed by Treventis for the treatment of Alzheimer’s disease and other neurodegenerative disorders characterized by protein misfolding.[3][9] Using Treventis’ proprietary Common Conformational Morphology (CCM) computational platform, TRV-101 was designed to bind selectively to misfolding-prone regions of both amyloid precursor protein–derived Aβ and tau, preventing formation of neurotoxic oligomers and aggregates in vitro and in animal models.[3][9][13] Preclinical data show that TRV-101 reduces toxic Aβ and tau aggregates, demonstrates favorable ADME properties, high brain penetrance, and oral bioavailability, and appears benign across a 44-receptor safety panel, supporting its development as a first-in-class dual Aβ/tau misfolding inhibitor for disease-modifying therapy in Alzheimer’s disease.[3][9][13]
