Overview

TRV734 is an orally bioavailable small molecule developed by Trevena for the treatment of opioid use disorder and as an analgesic for moderate-to-severe acute and chronic pain. It acts as a G-protein–biased agonist at the mu-opioid receptor, preferentially activating G-protein signaling while minimizing beta-arrestin recruitment. This mechanism is designed to provide effective analgesia with reduced adverse effects commonly associated with traditional opioids, such as constipation and respiratory depression. Preclinical and clinical studies have shown that TRV734 produces potent analgesic effects with less gastrointestinal dysfunction compared to morphine or oxycodone. The drug is being developed in collaboration with the National Institute on Drug Abuse (NIDA) for medication-assisted therapy in opioid use disorder[1][2][3][4][5][6][7][8].
