TRX-818M1

TRX-818M1 is an orally bioavailable small molecule inhibitor of the Hypoxia-Inducible Factor 1-alpha (HIF-1α) pathway, developed by the Swedish biotechnology company Trioxia. HIF-1α is a critical transcription factor that mediates the cellular response to hypoxia, a condition frequently found in the core of solid tumors. Under hypoxic conditions, HIF-1α promotes the expression of genes involved in angiogenesis, metabolic reprogramming (such as increased glycolysis), and cell survival, which collectively contribute to tumor progression and resistance to therapy. By inhibiting the HIF-1α pathway, TRX-818M1 aims to disrupt these adaptive mechanisms, thereby inhibiting tumor growth and potentially enhancing the efficacy of concurrent treatments. The compound has been evaluated in Phase 1 clinical trials for patients with advanced solid malignancies, including colorectal, breast, and pancreatic cancers.
