Overview

TRX518 is a humanized monoclonal antibody that targets the glucocorticoid-induced tumor necrosis factor receptor (GITR), a member of the TNF receptor superfamily expressed on various immune cells, including T regulatory cells and activated T effector cells. By binding to GITR and acting as an agonist, TRX518 enhances anti-tumor immune responses by activating tumor-antigen-specific T effector cells and reducing immunosuppression mediated by regulatory T cells within tumors. This mechanism aims to increase the elimination of cancerous cells and generate durable anti-tumor immunity. Developed initially by Tolerx and later advanced by Leap Therapeutics, TRX518 has been investigated in phase 1 clinical trials for multiple advanced solid tumors, including melanoma, prostate cancer, ovarian cancer, triple-negative breast cancer, and other unspecified solid tumors[1][2][4][5][6].
