TRX518 + pembrolizumab

TRX518 + pembrolizumab is a combination of two immunotherapy agents under investigation for the treatment of advanced solid tumors. TRX518 is a fully humanized monoclonal antibody that acts as an agonist of the glucocorticoid-induced TNF receptor-related protein (GITR), a member of the tumor necrosis factor receptor family expressed on T cells, B cells, and NK cells. By stimulating GITR, TRX518 enhances anti-tumor immune responses by reducing immunosuppressive regulatory T-cells and promoting effector T-cell activity[2][5][8]. Pembrolizumab is a monoclonal antibody that targets programmed cell death protein 1 (PD-1) on T cells, blocking its interaction with PD-L1/PD-L2 and thereby reinvigorating exhausted antitumor CD8+ T cells within the tumor microenvironment[2][3]. The rationale for combining these agents lies in their complementary mechanisms to enhance anti-tumor immunity. This combination has been evaluated in phase 1 clinical trials for patients with relapsed or refractory metastatic cancers who have not responded to standard therapies[2][6].
