Tryptanthrin-6-oxime (TRYP-Ox) is a synthetic oxime derivative of the natural alkaloid tryptanthrin, which is found in various plants such as *Polygonum tinctorium*. It is being investigated for its anti-inflammatory and therapeutic potential in autoimmune diseases, particularly rheumatoid arthritis (RA). The compound acts as a kinase inhibitor, specifically targeting the c-Jun N-terminal kinase (JNK) signaling pathway. By inhibiting JNK, tryptanthrin-6-oxime suppresses the expression and secretion of matrix metalloproteinases (MMP-1 and MMP-3) and various proinflammatory cytokines, including IL-6, IL-17A, GM-CSF, and RANKL. In preclinical murine models of arthritis, such as collagen-induced arthritis (CIA) and collagen-antibody-induced arthritis (CAIA), tryptanthrin-6-oxime has demonstrated efficacy in reducing clinical symptoms and joint damage comparable to or exceeding that of its parent compound, while exhibiting no significant cytotoxicity in vitro.
