Overview
The tryptophan-free amino acid drink (TFD) is an experimental research compound used as a challenge agent to induce acute tryptophan depletion (ATD). By providing a concentrated mixture of large neutral amino acids (LNAAs) while excluding tryptophan, the drink stimulates hepatic protein synthesis and creates intense competition for the LNAA transporter at the blood-brain barrier. This results in a rapid, temporary reduction in plasma and brain tryptophan levels, which is the essential precursor for serotonin (5-HT) biosynthesis. TFD is widely utilized in clinical research to investigate the role of the serotonergic system in mood regulation, cognitive function, sleep architecture, and the pathophysiology of psychiatric disorders such as major depressive disorder and alcohol dependence.
