Overview

TS-022 is a small molecule prostanoid DP1 receptor (prostaglandin D2 receptor, PTGDR) agonist developed by Taisho Pharmaceutical for the topical treatment of atopic dermatitis and pruritus. It acts by activating the prostanoid DP1 receptor, which leads to suppression of scratching behavior and acceleration of cutaneous barrier repair in preclinical models. The drug has demonstrated efficacy in reducing pruritus and improving skin inflammation in animal models of atopic dermatitis, with its effects being antagonized by a DP1 receptor antagonist. Its mechanism is distinct from corticosteroids or calcineurin inhibitors as it primarily targets itch and barrier repair rather than broad immunosuppression[1][2][4][6].
