TS-134 is an orally administered prodrug that is rapidly converted in the body to its active form, MGS0008. MGS0008 is a potent and selective agonist of metabotropic glutamate receptor 2 (mGluR2) and metabotropic glutamate receptor 3 (mGluR3). The drug has demonstrated antipsychotic-like activity in animal models by normalizing abnormal frontal cortical activity associated with NMDA receptor hypofunction—a pathophysiological feature relevant to schizophrenia. In clinical studies, TS-134 has shown good oral bioavailability and central nervous system penetration. It is being developed primarily for the treatment of schizophrenia by Taisho Pharmaceutical[1][3][4][5][8].
