TS-143 (also known as TP0463518) is a small molecule drug that acts as a competitive pan-inhibitor of prolyl hydroxylase domain enzymes 1, 2, and 3 (PHD1/2/3). By inhibiting these enzymes, TS-143 stabilizes hypoxia-inducible factor-2 alpha (HIF-2α), particularly in the liver. This stabilization leads to increased erythropoietin production and enhanced iron absorption via upregulation of intestinal iron transporters such as divalent metal transporter 1 and duodenal cytochrome b. The drug has demonstrated efficacy in preclinical models for improving anemia of inflammation without exacerbating liver inflammation. It is being developed primarily for the treatment of various forms of anemia, including acute posthemorrhagic anemia and anemia associated with chronic kidney disease or aging[1][3]. Clinical studies have evaluated its safety, pharmacokinetics, and pharmacodynamics in healthy adult males[2][5].
