The TSA-DC vaccine is an autologous, personalized dendritic cell-based cancer vaccine developed by BGI, China, for the treatment of advanced gastrointestinal solid tumors, including gastric and colorectal cancers. The therapeutic process involves identifying patient-specific tumor-specific antigens (neoantigens) through whole-exome and RNA sequencing of the patient's tumor tissue. These neoantigens, selected for their high predicted affinity for the patient's MHC molecules, are then loaded onto autologous dendritic cells derived from the patient's peripheral blood. Once administered subcutaneously, these mature dendritic cells present the neoantigens to the host immune system to elicit a targeted T-cell response against the tumor. Clinical protocols typically involve cyclophosphamide preconditioning to deplete regulatory T cells and enhance the vaccine's efficacy.
