TSB-IL17A is a preclinical-stage oligonucleotide therapeutic developed by TargetSite Therapeutics, a Yale University spinout. It utilizes the company's proprietary TargetSite Blocker (TSB) platform to modulate IL-17A expression. Unlike traditional antisense oligonucleotides that trigger RNase H-mediated cleavage, TSB-IL17A specifically blocks the binding of microRNA miR466l-3p to the 3' untranslated region (UTR) of IL-17A mRNA. In this specific biological context, miR466l-3p acts as an enhancer of IL-17A expression by cooperating with the RNA-stabilizing protein HuR; by disrupting this interaction, TSB-IL17A promotes mRNA decay and reduces IL-17A protein production. The drug is being investigated for the treatment of chronic inflammatory and autoimmune conditions, including autoimmune uveitis, psoriasis, and multiple sclerosis (modeled by experimental autoimmune encephalomyelitis).
