TSB-IL23A is a preclinical-stage oligonucleotide therapeutic being developed by TargetSite Therapeutics, a Yale University spinout. The drug utilizes the company's proprietary TargetSite Blocker (TSB) platform, which employs a novel mechanism of action to modulate gene expression. Specifically, TSB-IL23A is designed to inhibit or stabilize the interaction between microRNAs (miRNAs) and the messenger RNA (mRNA) of Interleukin-23 subunit alpha (IL-23A). By interfering with these unconventional miRNA-mediated mRNA stabilization pathways, the drug modulates the stability of IL23A mRNA, leading to a reduction in the expression of the pro-inflammatory cytokine IL-23A. This therapeutic approach is aimed at treating chronic inflammatory and autoimmune diseases where the IL-23 pathway is a validated driver of pathology.
