TSC-100 is a donor-derived T-cell receptor engineered T-cell therapy (TCR-T) designed to selectively eliminate residual disease cells post-hematopoietic cell transplantation (HCT) by targeting the minor histocompatibility antigen HA-1. This antigen is only expressed on hematopoietic cells of HLA-A*02:01-positive patients. TSC-100 is being developed by TScan Therapeutics for the treatment of hematologic malignancies including acute myeloid leukemia (AML), myelodysplastic syndromes (MDS), and precursor cell lymphoblastic leukemia-lymphoma. The therapy aims to eliminate all remaining blood cells in the host while ignoring the cells received from the donor after an allogeneic stem cell transplant, thereby preventing disease relapse. The manufacturing process involves isolation of peripheral blood mononuclear cells (PBMCs) from donor apheresis product and nucleofection with a transposon/transposase system delivered into pan T cells.
