TSC-100 + TSC-101 is an investigational combination of genetically engineered, donor-derived T cell receptor (TCR)-T cell therapies developed to prevent relapse of hematologic malignancies after allogeneic hematopoietic cell transplantation (HCT) in HLA-A*02:01 positive patients. Each component is a TCR-engineered T cell product: TSC-100 targets minor histocompatibility antigen HA-1, and TSC-101 targets HA-2—both antigens expressed on hematopoietic cells and presented by HLA-A*02:01. These therapies selectively eliminate residual host hematopoietic cells (including residual leukemia cells) without harming donor-derived cells. Administration follows HCT from haploidentical, matched unrelated, or mismatched unrelated donors. The products are manufactured by isolating donor PBMCs, transducing them to express antigen-specific TCRs, purifying and expanding the engineered T cells, and formulating them for infusion. They are administered intravenously after count recovery post-HCT. Initial trial data indicate favorable safety with no cytokine release syndrome or neurotoxicity and biological activity (donor chimerism rates, minimal residual disease suppression).
