TSC-101 is an engineered T cell therapy developed by TScan Therapeutics for the treatment of hematologic malignancies, including acute myeloid leukemia (AML), myelodysplastic syndromes (MDS), and precursor B-cell lymphoblastic leukemia-lymphoma. It is a form of adoptive cell therapy in which donor-derived T cells are genetically modified to express a T cell receptor (TCR) specific for the minor histocompatibility antigen HA-2, which is expressed on blood cells. The primary mechanism of action involves immunologic cytotoxicity—engineered T cells recognize and eliminate residual patient-derived hematopoietic cells after allogeneic hematopoietic cell transplantation (HCT), aiming to reduce relapse rates and increase relapse-free survival. The manufacturing process includes isolation of PBMCs from donors, genetic modification with a transposon/transposase vector encoding the HA-2-specific TCR, expansion, formulation, and cryopreservation before administration[4][5][6][8].
