Overview

TSC-200-A0201 is an autologous, enhanced T cell receptor-engineered T cell (TCR-T) therapy developed by TScan Therapeutics for the treatment of solid tumors associated with human papillomavirus type 16 (HPV16). The therapy utilizes a naturally occurring, high-avidity TCR specific for the HPV16 E7 antigen presented on HLA-A*02:01. Engineered patient-derived CD4+ and CD8+ T cells are modified to express this cancer antigen-specific TCR, along with co-expressed CD8α/β to enhance helper function and a dominant negative form of transforming growth factor beta receptor II (DN-TGFβRII) to promote persistence in the immunosuppressive tumor microenvironment. Preclinical studies demonstrated potent target-dependent cytotoxicity, cytokine secretion, proliferation, and anti-tumor efficacy in vitro and in vivo. The product is designed as part of a multiplexed approach (T-Plex), allowing combination with other engineered TCR-Ts targeting different antigens/HLA types based on individual tumor biology[6][7][8].
