TSC-201 is a T cell receptor engineered T cell (TCR-T) therapy program developed by TScan Therapeutics targeting melanoma-associated antigen C2 (MAGE-C2). The program includes multiple variants designed for different HLA types. **TSC-201-B0702** is engineered for HLA-B*07:02 presentation and includes a dominant-negative TGF-beta receptor II (DN-TGFβRII) to provide resistance to immunosuppressive tumor microenvironments. It comprises autologous T cells (both CD4+ and CD8+) engineered to express a therapeutic TCR that recognizes MAGE-C2 peptides presented on HLA-B*07:02. The therapy also expresses DHFRdm for methotrexate resistance, an exogenous CD8 co-receptor, and a CD34 epitope tag. The drug has been cleared by the FDA for clinical development and is being evaluated in Phase 1 trials as both monotherapy and in T-Plex combination regimens for various solid tumors including melanoma, anal cancer, cervical cancer, head and neck cancer, non-small cell lung cancer, and ovarian cancer. A separate variant **TSC-201 (HLA-A*02:01)** is in preclinical development.
