TSC-202-A0201 is an autologous, engineered T cell receptor (TCR)-T cell therapy targeting the melanoma-associated antigen 4 (MAGEA4) presented by HLA-A*02:01. Developed by TScan Therapeutics, this therapy is designed to recognize and eliminate cancer cells expressing MAGEA4 in the context of HLA-A*02:01. The mechanism of action involves genetically modifying a patient's own T cells to express a high-affinity, MAGEA4-specific TCR, enabling targeted immunologic cytotoxicity against tumor cells. Preclinical studies have demonstrated specificity and potency with no observed alloreactivity across 110 common HLA-I allotypes. The product has advanced into Phase 1 clinical trials for multiple solid tumors including anus neoplasms, head and neck neoplasms, non-small cell lung cancer, sarcoma, and uterine cervical cancer[1][2][3].
