TSC-203-A0201 + TSC-202-A0201 is a combination of two investigational autologous, engineered T cell receptor (TCR)-T cell therapies developed by TScan Therapeutics for the treatment of solid tumors. Each component targets a different tumor-associated antigen presented on HLA type A*02:01. TSC-203-A0201 targets Preferentially Expressed Antigen in Melanoma (PRAME), while TSC-202-A0201 targets Melanoma-associated antigen 4 (MAGEA4). Both are designed to recognize and kill cancer cells expressing these antigens via immunologic cytotoxicity. The therapies use patient-derived (autologous) CD8+ and/or CD4+ lymphocytes genetically modified to express specific high-affinity, cancer-targeting TCRs, with enhancements such as co-delivery of CD8α/β coreceptors and dominant-negative transforming growth factor beta receptor II (DN-TGFβRII) to improve efficacy and persistence[2][3][5]. The combination is being evaluated in Phase 1 clinical trials as part of the "T-Plex" platform for patients with various solid tumors including head and neck neoplasms, non-small cell lung cancer, sarcoma, uterine cervical cancer, anal neoplasms, among others[3][7]. The approach aims to address tumor heterogeneity by targeting multiple antigens simultaneously.
