TSC-204 is an investigational, autologous T-cell therapy in which patient-derived T cells are genetically engineered to express a T-cell receptor (TCR) that recognizes melanoma-associated antigen 1 (MAGE-A1) presented by specific human leukocyte antigen (HLA) alleles (most notably HLA-A*02:01 and HLA-C*07:02)[1][4][6][13]. Upon administration, these engineered T cells specifically bind to and target MAGE-A1-expressing tumor cells, resulting in immunologic cytotoxicity and T lymphocyte replacement in the tumor[4][13]. TSC-204 is developed using TScan Therapeutics’ platform and is currently being evaluated in Phase 1 clinical trials for multiple solid tumors, including non-small cell lung cancer, head and neck cancer, anal cancer, cervical cancer, ovarian cancer, melanoma, and other advanced malignancies[1][4][5][13]. The therapy is being investigated both as a single agent and in combination regimens (T-Plex) with other engineered T-cell products targeting distinct antigens/HLA alleles[2][8][12].
