Overview

TSC-204-A0101 is an autologous, engineered T cell receptor (TCR)-T cell therapy developed for the treatment of solid tumors expressing the cancer-testis antigen MAGE-A1 in patients with HLA-A*01:01. The therapy uses a naturally occurring TCR identified via TScan’s ReceptorScan platform and is further enhanced by co-expression of CD8α and CD8β coreceptors to improve helper T cell function, as well as a dominant-negative transforming growth factor beta receptor II (DN-TGFβRII) to increase resistance to immunosuppressive signals in the tumor microenvironment. The product is manufactured using a transposon-based vector encoding these elements along with a mutated dihydrofolate reductase for selection. Preclinical studies have demonstrated target-dependent cytotoxicity, proliferation, cytokine secretion, and anti-tumor efficacy. It is being evaluated in Phase 1 clinical trials for multiple solid tumor indications[6][2][3].
