TSC-204-A0201 + TSC-201-B0702 is a combination of two investigational autologous, engineered T cell receptor (TCR)-T cell therapies developed by TScan Therapeutics for the treatment of solid tumors. Each component targets a specific cancer-associated antigen presented by defined HLA types: - **TSC-204-A0201** targets Melanoma-associated antigen 1 (MAGE-A1) on HLA-A*02:01-positive tumor cells. - **TSC-201-B0702** targets Melanoma-associated antigen C2 (MAGE-C2) on HLA-B*07:02-positive tumor cells[1][6][8]. Both are part of the "T-Plex" platform, which enables multiplexed targeting to address tumor heterogeneity and potential immune escape due to loss of individual antigens or HLA alleles[5][6]. The therapies are being evaluated in a Phase 1 clinical trial for multiple solid tumors, including HPV-related cancers and melanoma. The mechanism involves engineering patient-derived (autologous) T cells with high-affinity, antigen-specific TCRs that recognize and kill cancer cells expressing the target antigens in the context of their respective HLAs[8].
