TSC-204-C0702 + TSC-201-B0702 is a combination of two investigational autologous, engineered T cell receptor (TCR-T) therapies developed by TScan Therapeutics for the treatment of solid tumors. Each component targets a distinct cancer-associated antigen presented by specific HLA types: - **TSC-204-C0702** targets Melanoma-associated antigen A1 on HLA-C*07:02. - **TSC-201-B0702** targets Melanoma-associated antigen C2 on HLA-B*07:02[1][6]. These therapies are designed to recognize and eliminate tumor cells expressing their respective antigens, potentially overcoming tumor heterogeneity and immune evasion due to HLA loss. The combination is being evaluated in the Phase 1 "T-Plex" clinical trial (NCT05973487), which investigates multiplexed administration of different TCR-Ts in patients with advanced solid tumors such as melanoma, non-small cell lung cancer, head and neck cancers, cervical cancer, anal cancer, vulvar squamous cell carcinoma, penile squamous cell carcinoma, and other HPV-related or MAGE-expressing malignancies[5][7]. The products are manufactured using a nonviral transposon/transposase system that enables rapid engineering of both cytotoxic and helper CD8+ and CD4+ T cells for enhanced anti-tumor activity[8].
