TSC-303-A02 is an investigational, in vivo-engineered T-cell receptor (TCR)-T cell therapy being developed by TScan Therapeutics for the treatment of solid tumors, with a particular focus on PRAME-expressing malignancies such as cutaneous melanoma. Unlike conventional autologous TCR-T therapies that require complex and costly ex vivo manufacturing, TSC-303-A02 utilizes a T-cell-targeted lentiviral vector to deliver a TCR gene directly to T cells inside the patient's body. This TCR is designed to specifically recognize and bind the cancer-testis antigen PRAME (preferentially expressed antigen in melanoma) presented on HLA-A*02:01 molecules. By engineering T cells in vivo, the therapy aims to eliminate the need for patient-specific manufacturing, reduce vein-to-vein time, and bypass the requirement for lymphodepleting chemotherapy. TSC-303-A02 is currently in IND-enabling preclinical development.
