Drug intelligence / Profile preview
TSD-IL-7 is an engineered interleukin-7 (IL-7) cytokine fused to a tissue transglutaminase 2 (TG2) substrate domain. It is designed to undergo TG2-mediated covalent crosslinking to lysine-containing extracellular matrix (ECM) components specifically within the tumor microenvironment, where TG2 enzymatic activity is elevated. This localized anchoring enhances the retention and bioactivity of systemically administered IL-7, promoting T cell proliferation, reducing T cell exhaustion, and stimulating CD8+ central memory T cell development. Preclinically, TSD-IL-7 has demonstrated potent antitumor activity across multiple solid tumor models, including ovarian cancer, both as a monotherapy and in combination with anti-PD-1 immune checkpoint inhibitors, without inducing systemic toxicity.
