TSG-02 is a recombinant humanized fusion protein developed as a biosimilar to aflibercept. It functions as a "VEGF trap" by incorporating the second Ig domain of human VEGF receptor 1 (VEGFR-1) and the third Ig domain of human VEGF receptor 2 (VEGFR-2) fused to the Fc portion of human IgG1. This structure allows TSG-02 to bind with high affinity to all isoforms of Vascular Endothelial Growth Factor A (VEGF-A), as well as VEGF-B and Placental Growth Factor (PlGF). By sequestering these growth factors, TSG-02 inhibits the activation of VEGF receptors, thereby suppressing pathological neovascularization and reducing vascular leakage in the eye. Developed by Shanghai Teruisi Pharmaceutical and Shanghai Huiyu Biotech, the drug is primarily investigated for the treatment of retinal vascular diseases, including diabetic macular edema (DME) and neovascular age-related macular degeneration (nAMD).
