TSG6dHep-Fc is a recombinant fusion protein developed by Halozyme Therapeutics as a diagnostic imaging agent to detect hyaluronan (HA) accumulation in solid tumors. It consists of a fragment of the tumor necrosis factor-stimulated gene-6 (TSG6) protein, which has been mutated at its heparin-binding site (dHep) to reduce non-specific binding, fused to a human IgG1 Fc domain. TSG6dHep-Fc acts as a soluble HA-binding protein that can be labeled with imaging agents, such as the near-infrared fluorophore DyLight755, to non-invasively identify HA-rich malignancies. This tool was designed to facilitate patient selection for HA-targeting therapies, such as pegylated recombinant human hyaluronidase (PEGPH20), by confirming the presence of high HA levels in the tumor extracellular matrix and monitoring enzymatic activity.
