Overview

TSHR-CAR-T is an experimental autologous **CAR-T cell therapy** designed for Graves' disease and potentially other antibody-mediated autoimmune thyroid diseases. The construct uses the extracellular domain of the thyroid-stimulating hormone receptor as the antigen-recognition moiety, linked to a CD8 transmembrane region and a 4-1BB costimulatory signaling domain, to redirect T cells against autoreactive B lymphocytes that produce thyroid-stimulating hormone receptor antibodies. The intended mechanism is selective depletion of TRAb-producing pathogenic B cells in order to reduce the autoimmune drive underlying hyperthyroid Graves' disease. Available information supports a preclinical-stage biologic/cell-therapy program, and no specific sponsoring company, trade name, or clinical-stage registration was identified from the provided context.
