TSHR targeting CAR-T cell therapy is an investigational cellular immunotherapy designed to treat advanced thyroid cancers by engineering a patient’s own T cells to express a chimeric antigen receptor (CAR) that specifically recognizes the thyrotropin (thyroid-stimulating hormone) receptor (TSHR). The mechanism of action involves genetically modifying autologous or allogeneic T cells with a lentiviral vector encoding a single-chain variable fragment (scFv) derived from monoclonal antibodies against human TSHR. These engineered cells are equipped with co-stimulatory domains such as CD28 or 4-1BB and the CD3ζ signaling domain to enhance activation and persistence. Upon infusion, these CAR-T cells selectively bind to and kill tumor cells expressing high levels of TSHR through cytotoxic mechanisms including cytokine release and granzyme-mediated apoptosis. Preclinical studies have demonstrated potent anti-tumor efficacy in vitro and in vivo models of differentiated thyroid cancer (DTC), with early-phase clinical trials indicating promising safety and efficacy profiles for relapsed/refractory thyroid cancer[1][2][4][5].
