Overview

TSL-101 is an inhaled small molecule being developed by Tessel Biosciences for the treatment of muco-obstructive lung diseases, including cystic fibrosis, chronic obstructive pulmonary disease (COPD), and bronchiectasis. It functions as a potent and durable inhibitor of the epithelial sodium channel (ENaC). By blocking ENaC, TSL-101 prevents the excessive absorption of sodium and water from the airway surface, thereby increasing airway surface liquid (ASL) hydration. This rehydration helps to normalize mucus viscosity and improve mucociliary clearance, addressing the underlying pathophysiology of mucus stasis and airway obstruction. The drug is currently in preclinical development and is designed for localized delivery to the lungs via inhalation to minimize systemic exposure.
