TSLP siRNA is a therapeutic modality consisting of small interfering RNA molecules designed to specifically target and silence the expression of the Thymic Stromal Lymphopoietin (TSLP) gene. TSLP is an epithelial-derived cytokine (alarmin) that acts as a master regulator of type 2 (T2) immune responses, making it a key therapeutic target for inflammatory airway diseases. By utilizing the RNA interference (RNAi) pathway, TSLP siRNA triggers the degradation of TSLP mRNA within pulmonary cells, thereby preventing the synthesis and secretion of the TSLP protein. This reduction in TSLP levels limits the activation of dendritic cells and the subsequent induction of Th2-driven allergic inflammation. Developed by companies such as Arrowhead Pharmaceuticals (e.g., ARO-TSLP), these siRNAs are often engineered for lung-targeted delivery (e.g., via inhalation) and may be formulated as monomeric agents or as part of multi-targeted dimer platforms that simultaneously silence other alarmins like IL-33 to achieve broader anti-inflammatory effects in conditions such as asthma and chronic obstructive pulmonary disease (COPD).
