Overview

TSN222 is a novel bifunctional small molecule drug developed as a dual-action tumor immune agonist. It is designed to sequentially release both immune agonism and cytotoxic activity, modulated by its pharmacokinetics. Mechanistically, TSN222 acts as both a stimulator of interferon genes (STING) agonist and an inhibitor of prolyl 4-hydroxylase subunit beta (P4HB). Upon administration, it activates interferon production within the tumor microenvironment to stimulate innate immunity. Subsequently, it is metabolized in vivo into a DNA synthesis inhibitor that directly kills tumor cells, providing antigens and DNA to further prime the adaptive immune system and enhance STING pathway activation synergistically. Preclinical data have shown good tolerability, robust tumor suppression effects, and durable immune memory responses. The drug is currently being evaluated in first-in-human phase 1/2 clinical trials for advanced solid tumors and lymphomas[2][3][6].
