**tspan8-specific car t cells** are a form of chimeric antigen receptor T cell therapy engineered to selectively recognize and kill cells expressing TSPAN8, a tetraspanin overexpressed on the surface of various epithelial and solid tumor cancer cells, including pancreatic ductal adenocarcinoma. The therapy involves extracting a patient’s T cells, genetically engineering them to express a CAR construct that specifically binds to TSPAN8, and re-infusing these cells back into the patient to mount a targeted immune response against TSPAN8-expressing tumor cells. Preclinical studies demonstrated significant anti-tumor efficacy both in vitro and in mouse xenograft models, with varying degrees of tumor clearance and disease stabilization depending on CAR design and target expression. TSPAN8 is considered a promising target due to its cancer-specific upregulation, but safety concerns remain, especially regarding potential on-target, off-tumor toxicity in healthy gastrointestinal tissues where TSPAN8 is also expressed. The primary indication is for cancers such as pancreatic ductal adenocarcinoma, but broad applicability to other epithelial cancers may exist[1][2][3].
