TSPO siRNA is a small interfering RNA designed to silence the expression of the 18 kDa translocator protein (TSPO), also known as the peripheral benzodiazepine receptor. TSPO is an outer mitochondrial membrane protein primarily involved in cholesterol transport and neurosteroid synthesis. In research settings, TSPO siRNA is utilized as a tool to investigate the physiological role of TSPO in various cellular processes, such as airway smooth muscle (ASM) cell contraction, growth, and migration. By achieving significant knockdown of TSPO expression (typically around 80%), researchers can assess the impact of TSPO deficiency on mitochondrial structural features, membrane potential, and intracellular calcium flux. Studies have indicated that while TSPO knockdown may not significantly alter baseline ASM cell stiffness or histamine-induced contraction, it serves as a critical control for evaluating the specific actions of TSPO ligands.
