Overview

TSR-825 (also known as GSK4069825) is a bispecific monoclonal antibody designed to simultaneously target and inhibit two immune checkpoints: Programmed Cell Death Protein 1 (PD-1) and T-cell Immunoglobulin and Mucin-domain containing-3 (TIM-3). Developed by Tesaro (a subsidiary of GSK), the drug aims to overcome resistance to PD-1/PD-L1 monotherapy by blocking TIM-3, which is often co-expressed with PD-1 on exhausted T cells in the tumor microenvironment. By dual inhibition, TSR-825 is intended to enhance T-cell activation and anti-tumor immune responses more effectively than single-agent checkpoint inhibitors. It was primarily investigated in Phase 1/2 clinical trials for the treatment of advanced solid tumors before being removed from the GSK development pipeline for strategic reasons.
